First, make sure the default profile is pointing to the proper location of the templates and printers and then save it. Are your templates and printers on a network share...like AraxiPreps? If so you might want to make sure the network drive is mounted on the Desktop before trying to open Preps from Prinergy. Although Preps should try to mount the drive if launched from Prinergy I've seen it have problems doing it. Have also seen it not see the volume even if it is mounted if it was mounted via the IP address. Mounting it by the computer name of the volume works but not the IP address in some cases.
